This is a method by which microorganisms from a liquid culture can be transferred to a tube of sterile medium and is intended to supplement the instruction of the technique in the general (non-medical) microbiology course in which I am the instructor. "Aseptic technique" prevents cross contamination of the culture and the environment. For introductory microbiology labs where potential or actual pathogenicity is not an issue, this method is sufficient - keeping in mind the necessity of carefully flaming the loop from the base of the wire to the tip when the process is completed in order to prevent an aerosol. Many courses do not teach this! (Also wiping with work area with disinfectant before and after - and good washing of the hands.) One may need to utilize personal protective equipment ("PPE") and/or work in a safety hood (provided with laminar airflow) depending on the Hazard Group to which particular species are assigned. Naturally we wouldn't think of performing the procedure as described for something like the cholera organism! The oral presentation is provided by the Apple Macintosh OS 9 "Victoria, high quality" voice which (in my humble opinion) is far superior in clarity and pronunciation to its OS X counterpart. Certainly better than mine.
